Genomic DNA (~ 100 ng) isolated from mouse tails cut from WT(+/+), heterozygous (+/-) or knock-out mouse(-/-) is tested for genotyping PCR. A leading brand A and GX DNA Taq polymerase (1 unit) are used in a 20 µl reaction system for 35 cycles. Lane 10: previously defined WT and KO mouse genomic DNA as positive control.
Concentration 5 units/µl, in 50 mM Tris HCl (pH 7.9), 50mM KCl, 0.1 mM EDTA (pH 8.0), 1 mM DTT, 0.5% Tween-20 (v/v), and 50% glycerol (v/v).
10 X Reaction Buffer (with Mg++) 100 mM Tris HCl (pH 8.5), 500 mM KCl, 20 mM MgCl2.
Storage -20 °C.
Unit definition One unit incorporates 10 nmol of deoxyribonucleotide into acid perceptible material in 30' at 74 °C.
